Investigation of mRNA Levels of PFK-1, LDH-A, p53, and HIF-1α Genes Involved in Metabolic Reprogramming in Non-small Cell Lung Carcinoma

Background: It is expected that the expression of key enzymes glycolysis pathway, specially PFK-1, increases tumor cells and so enhances function this pathway. The p53 HIF-1 proteins are regulators PFK-1 LDH enzymes. This study was performed to investigate changes in LDH-A, p53, HIF-1α genes identify metabolic non-small cell lung carcinoma (NSCLC) samples. Methods: A number 30 tumors their adjacent normal tissue samples from surgically approved NSCLC patients were used. Total RNA each extracted. mRNA levels PFK-1M, LDHA, hypoxia-inducible factor-1 (HIF-1α) evaluated tissues all using real-time polymerase chain reaction (PCR) method. Finally, statistical analysis used determine significant differences relationship between levels. Results: According results, there no difference these tissues. level lactate dehydrogenase (LDHA) adenocarcinoma (AdC) squamous (SqCC) types observed (P=0.014). Also, LDHA metastatic non-metastatic (P=0.035 P=0.046 respectively). Age male gender directly associated with an increased risk NSCLC. Conclusion: results present revealed increase may be involved initiation, LDH-A HIF-1a a phenotype.
